Brain tumours (BT) patients treated with radio- and chemotherapy may develop a second primary malignancy. It is known that receptor tyrosine kinases (RTKs) initiate a signal transduction leading to uncontrolled cell growth and to cytoprotection against cancer treatment. RTKs have recently been identified that suitable brain cancer markers and therapeutically targets. In our previously studies, we found that inhibition of RTKs by small molecular inhibitors, induced cytotoxicity and increased radiosensitivity in glioblastoma (GB) cells. We also found that Helianthine has anticancer activity on human GB cells. To our knowledge, this is the first report indicating that Helianthine induces cell death in GB cells by downregulation the RTKs activity. Since 2006, we have collected 250 brain cancer samples from patients that were operated in the Dep. of Neurosurgery, "Bagdasar-Arseni" Hospital, Bucharest, and developed a Brain Tumour Bank (BTB). Usually, large numbers of samples must be collected to provide researchers enough material studies. In light of these considerations, the aims of this study are: 1.To analyse the general nature of the cytotoxic effect of Helianthine on BT cells 2.To develop in vitro screening models for the optimally effective combination regimes, from which in vivo programs can be made to test several receptor protein tyrosine kinases inhibitors (RPKIs) 3.To sustain and expand en existing BTB that contains specimens from patients with cerebral tumours.
